摘 要:为优化板栗壳黄酮类色素的提取工艺并对其稳定性、抗氧化性及抑菌能力进行研究,以超声波处理时间、料液比、乙醇体积分数、提取温度为优化因素,黄酮类色素产率为评价指标,通过单因素试验和响应面法优化板栗壳黄酮类色素的提取工艺,并对黄酮类色素的热稳定性、抗氧化性能和抑菌性能进行评价。结果表明:最佳提取工艺条件为:超声波处理时间36 min、料液比1∶13 g/mL、乙醇体积分数59%、提取温度54℃,此条件下板栗壳中黄酮类色素的产率为7.083%。热稳定结果显示,黄酮类色素在80℃以上耐热性较差。抗氧化结果显示黄酮类色素对1,1-二苯基-2-三硝基苯肼(DPPH)自由基和羟基自由基的清除率最高可达85.5%和67.4%,具有一定的抗氧化能力,但是其抗氧化能力比维生素C弱。通过滤纸片扩散法,发现板栗壳黄酮类色素对大肠杆菌和金黄色葡萄球菌的生长均具有一定的抑制作用。In order to optimize the extraction process of flavonoid pigments from chestnut shell and study its stability,antioxidant and bacteriostatic ability.With ultrasonic time,solid-liquid ratio,ethanol concentration and extraction temperature as optimization factors and flavonoid pigment yield as evaluation index,the extraction process of flavonoid pigment was optimized by single factor and response surface method,and the thermal stability,antioxidant and antibacterial properties of flavonoid pigment were evaluated.The results show that the optimal extraction conditions were ultrasonic time of 36 min,solid-liquid ratio of 1∶13 g/mL,ethanol concentration of 59%and extraction temperature of 54℃.Under these conditions,the yield of flavonoid pigments in chestnut shell was 7.083%.The thermal stability results show that the flavonoid pigments had poor heat resistance above 80℃.The antioxidant results indicated that flavonoid pigments exhibited a maximum scavenging rate of 85.5%and 67.4%against DPPH radicals and hydroxyl radicals,respectively,demonstrating certain antioxidant capacity,but were weaker than that of vitamin C.Through the filter paper diffusion method,it was found that the flavonoid pigment of chestnut shell had certain inhibitory effect on the growth of Escherichia coli and Staphylococcus aureus.
