摘  要:本研究旨在优化酒糟中纤维素酶酶促反应条件,对CMC-Na法测定酒糟中纤维素酶活力的影响因素进行探讨,建立了酒糟中纤维素酶活力的测定方法,并通过单因素试验和响应面法确定酒糟中纤维素酶酶促反应的最佳条件。结果表明,反应最佳条件为缓冲液pH值4.5、CMC-Na浓度4.85 mg/mL、反应温度48.96℃,此条件下酒糟中纤维素酶活力最高,为12.914U/g。实验结果可为后续酒糟中纤维素酶的研究奠定基础。The aim of the experiment was to optimize the enzymatic reaction conditions of cellulase in distillers grains,to explore the factors affecting the determination of cellulase activity in distillers grains by CMC-Na method,and to establish a determination method for cellulase activity in distill-ers grains.According to the results of single factor tests and response surface method,the optimal conditions for the enzymatic reaction of cellulase in distillers grains were as follows:buffer pH value 4.5,CMC-Na concentration 4.85 mg/mL,reaction temperature 48.96℃.Under these conditions,cel-lulase activity in distillers grains was the highest,reaching 12.914 U/g.The results have laid a foundation for further research on cellulase in distillers grains.
