摘 要:本研究针对食用菌及食用菌制品中的麦角甾醇,建立了一种基于液相色谱法(Liquid Chromatography,LC)的检测方法。样品经皂化、甲醇超声提取后,采用LC对样品溶液进行分析,以甲醇为流动相,紫外检测器于282 nm波长处检测,外标法定量。对所建立的方法进行分析性能的考察,麦角甾醇在0.5~200 mg/L范围内线性关系良好,在食用菌干制品中麦角甾醇的检出限为10.0 mg/kg,定量限为25.0 mg/kg,在鲜食用菌、食用菌罐头及食用菌酱制品中麦角甾醇的检出限为2.50 mg/kg,定量限为6.25 mg/kg。采用该方法对24种食用菌及其制品中的麦角甾醇进行分析,样品中的麦角甾醇在80.9~4506 mg/kg之间。采用加标回收试验验证方法的准确性,在24种食用菌及其制品中,麦角甾醇的回收率在80.1%~120%之间,精密度为2.31%~13.3%。结果表明,所建立的方法简便、易于批量操作且具有良好的准确性,可满足食用菌及食用菌制品中麦角甾醇的测定需求。A method based on liquid chromatography(LC)has been established for the determination of ergosterol in edible mushrooms and edible mushroom products.After saponification and ultrasonic extraction with methanol,the sample solution was introduced into the LC system for analysis.Methanol was used as the mobile phase,and the ultraviolet detector was set at a wavelength of 282 nm for detection,the sample solution was quantified using external an standard.The analytical performance of the established method was evaluated.Ergosterol exhibited a good linear relationship within the range of 0.5-200 mg/L.The limit of detection(LOD)for ergosterol in dried edible mushrooms was 10.0 mg/kg,and the limit of quantitation(LOQ)was 25.0 mg/kg.For fresh edible mushrooms,canned mushrooms,and mushroom sauce,the LOD for ergosterol was 2.50 mg/kg,and the LOQ was 6.25 mg/kg.This method was applied to analyze ergosterol in 24 types of edible mushrooms and their products,and the ergosterol concentrations ranged from 80.9 to 4506 mg/kg in the samples.The accuracy of the developed method was verified by spiking recovery experiments,and the recovery rates of ergosterol in the 24 types of edible mushrooms and their products ranged from 80.1%to 120%,with a precision of 2.31%to 13.3%.The results indicated that the established method is simple and suitable for batch operation and has good accuracy,meeting the requirements for the determination of ergosterol in edible mushrooms and their products.
