摘  要:本研究用生何首乌作为主要原料,以虫草与何首乌主要有效成分虫草素、甘露醇、二苯乙烯苷、游离蒽醌和游离蒽醌/总蒽醌比值为指标,从本实验室分离保存的5株虫草真菌中,筛选出一株既能较高生产虫草有效成分,又能有效降解生首乌中结合蒽醌的虫草真菌CS2。采用响应面法并结合数学规划模型进行培养基优化,得到CS2菌株给定模型的最佳培养基组分为:生首乌55.76%、葡萄糖2.56%、牛肉膏1.47%、麸皮34.24%、豆饼粉10%、酵母粉1%、蛋白胨1%,磷酸二氢钾0.2%,硫酸镁0.1%,氯化锌0.2%,发酵培养基含水量为:95m L/100g干基质。以此培养基,250m L三角瓶装40g湿培养基,25℃发酵培养6d,获得发酵产品中含虫草素396.27μg/g,甘露醇17.61mg/g,二苯乙烯苷6.04mg/g,游离蒽醌提高31.6%,结合蒽醌降解90.0%。The condition of the fermentation of Cordyceps fungus in Polygonum multiflorum as main raw material by response surface method and FNLP were studied. After choosing the most suitable Paecilomyces hepialid strain CS2 and on the basis of statistics principles, three factors at three levels response surface methodology was applied to build the model. The degree of overall effect was used as response value. By analyzing the response surface, the optimal culture medium was obtained as the follows: Polygonum multiflorum 55.76%, glucose 2.56%, beef extract1.47%, bran 34.24%, soybean cake powder 10%, yeast powder 1%, peptone 1%, KH2PO 40.2%, Mg SO40.1%,Zn Cl20.2% and water 95 m L/100 g dry weight. And after full factorial experiment, the most suitable production process is sample loading 40 g wet weight in conical flask(250m L), fermentation time 6d. And the composition of the production is cordycepin 396.27μg/g, mannitol 17.61mg/g, diphenyl ethylene 6.04mg/g, free anthraquinones increase by 31.6%, and combined anthraquinones decrease by 90.0%.
