摘 要:由出芽短杆霉发酵产生的普鲁兰多糖,发酵过程中会产生大量色素、蛋白质以及一些小分子糖类物质,增加了多糖提取难度。该实验将从发酵液除杂、脱色、去蛋白、小分子多糖出发,对后提取过程中用到的助滤剂硅藻土,脱色所需的活性炭,树脂选型,超滤及干燥方式的工艺参数进行优化。试验结果表明,选取一号硅藻土作为助滤剂,用量2%,滤液澄清度高、过滤速度快;脱色活性炭的添加量为0.5%、温度50℃、时间40 min、p H5.5,脱色效果好、普鲁兰多糖得率高;使用树脂型号D,二次脱色效果良好,同时能去除一些杂蛋白;超滤3次,单二寡糖的去除率能达到96.6%,采用滚筒干燥方式,干燥成本低,成品质量佳。In the process of pullulan polysaccharides fermentation,a large amount of by-products such as pigment,protein and small molecular sugars were produced at the same time,resulting in difficult extraction of polysaccharide. Thus,in this work,the separation of fermentation liquid,decoloration,remove of proteins and small molecular sugars were studied. The extraction conditions were optimized. The results showed that using diatomite 1 of 2% as filter aid,the clarity of filtrate was higher and the filtration was faster. It obtain a good decolorizing effect and high yield of pullulan polysaccharides when 0. 5% activated carbon was added at 50 ℃,p H 5. 5 for 40 min. Resin type D was used for secondary decolorizing and exhibited a good effect and could remove some miscellaneous protein. The removal rate of Mono-,di-,and oligosaccharides after ultra-filtration for three times could be up to 96. 6%. The product dried by drum drying method with low cost had good quality.
