摘 要:目的 :研究氧化苦参碱对不同状态下的TNFα、IL 6和IL 8的影响。方法 :利用脂多糖 (lipopolysaccharide,LPS)刺激小鼠腹腔巨噬细胞和人早幼粒白血病细胞作为过度炎症反应的体外模型 ,用角叉菜胶引起的小鼠足肿胀作为炎症反应的体内模型 ,采用生物学方法和放射免疫分析法测定体外细胞培养上清液和局部炎症渗出液中TNFα、IL 6和IL 8。结果 :氧化苦参碱对体外细胞培养上清液和局部炎症渗出液中TNFα、IL 6和IL 8均具有一定的抑制作用。结论 :氧化苦参碱的抗炎免疫作用可能与其抑制早期致炎性细胞因子的产生有关。Objective: To study the effect of oxymatrine on TNFα, IL 6 and IL 8. Methods: The TNFα, IL 6 and IL 8 in supernatant of culture cell and tissue exudate were respectively measured by methods of MTT and radioimmunoassay, in which lipopolysaccharide stimulation on mouse peritoneal macrophages and HL 60 cells were used as the external model of excess inflammation, and carrageenin induced mice ped was used as the internal model of inflammation. Results: Oxymatrine can inhibit TNFα, IL 6 and IL 8 in supernatant of culture cell and tissue exudate respectively. Conclusion: The mechanism of anti inflammation of oxymatrine is connected with its inhibition the production of cytokines (TNFα, IL 6 and IL 8).
