摘  要:用截留分子量2万的中空纤维膜超滤阿魏酸酯酶与阿拉伯木聚糖酶的混合酶制剂。研究了超滤进口压力、超滤温度对膜通量和酶活的影响,确定了进口压力0.10~0.15MPa、超滤温度25~30℃为最佳操作工艺参数。酶的浓缩倍数可达7倍以上,使酶制剂更加方便保存。采用2次超滤的方法可以去除浓缩酶液中90%的低聚糖和阿魏酸等小分子物质,解决了酶制剂使用中的产物抑制问题。Hollow fiber membrane with molecular cutoff weight of 20,000 was applied to concentrate ferulic acid esterase and arabioxylanase. The effect of ultrafiltration pressure and temperature on membrane flux and enzyme activities was studied. The optimal parameters of ultrafiltration were: pressure 0.10-0.15MPa, and temperature 25-30%. Under these conditions, the enzymes were concentrated by more than 7 times. Furthermore, 90% of oligosaccharides and ferulic acid in the concentrated enzymes was removed if the concentrates was diluted by 10 times and then ultrafiltrated again, thus reduced the substrate inhibition.
