摘  要:以糖化酵母和酿酒酵母为亲本,采用单亲灭活原生质体融合技术进行属间融合,同时对酵母原生质体形成和再生条件进行了探索,取培养6h ̄8h的单倍体在30℃下,2.0%的蜗牛酶酶解1.0h时,其形成率超过92%。并对双亲与融合子的细胞大小、DNA含量、淀粉利用率及产酒精能力等进行了比较研究;获得1株融合子,利用可溶性淀粉发酵,酒度可达6.5%(v/v)。A new fusant was constructed using the single parent killing protoplast fusion technology and S. diastaticus and S. cerevisiae as parents. The formation and regeneration conditions of yeast haploidy protoplast were also studied. The formation rate reached over 92.0% when culturing haploidy at 30% for 6h-8 h and analyzing the cell wall using 2.0% Snaillase for 1.0 h. The cell size, DNA content, starch utility rate and ethanol production ability between parents and fusant were also compared. Finaly, a fusant that could produce 6.5% (v/v) ethanol was obtained using soluble starch fermentation.
