摘 要:以形态特征为依据,对具有降解胆固醇能力的霉菌TS406菌株进行鉴定,在此基础上,通过单因素实验,以胆固醇氧化酶活性和胆固醇降解率为指标,确定该菌株发酵培养的最适碳、氮源及其比例,并对发酵条件进行初步研究。结果表明,霉菌TS406属于青霉属真菌,其发酵培养的最适碳、氮源分别是淀粉和酵母粉,两者在培养基中的最适浓度均为1.5g/L。发酵培养的最佳条件是:培养液的初始pH为6.5,培养温度为28℃,接种量为3%,装液量为30mL/250L,胆固醇氧化酶的活性可以达到1420U/L,发酵培养2d后胆固醇的降解率为91.2%。Mold TS406 was identified based on the morphological characteristics, the activity of cholesterol oxidase and degrading rate of cholesterol in liquid medium. The suitable carbon source, nitrogen source, the ratio of carbon to nitrogen and the fermented conditions were determined by the experiments of single factor. The results showed that the mold TS406 belonged to genus Penicillurn. The fermented conditions which are good for the activity of cholesterol oxidase and degrading rate of cholesterol were initial pH6.5 of medium, 28℃ culturing temperature, 3% inoculating volum and 30mL liquid medium in 250mL flask. The high- est enzyme activity is 1420U/L, and the degrading rate of cholesterol in medium is 91.2% after 2d shaking fermentation, when the medium contain 1.5g/L starch and yeast extract respectively.
