摘 要:采用单因素实验和正交实验研究了时间、温度、pH值、酶加量四个因素对提取川芎多糖的影响。单因素实验确定了纤维素酶、复合果胶酶提取川芎多糖的最佳工艺条件:纤维素酶0.15%,复合果胶酶6%,pH值3.5,时间150 min,温度60℃;正交实验得到川芎多糖的最佳工艺条件为:纤维素酶0.15%,复合果胶酶10%,时间210min,pH值3.5,温度60℃,在此条件下,多糖的提取率为3.03%。酶法提取川芎多糖简单、快捷,提取率较高。The influence factors including reaction time,pH, the added enzyme and reaction temperature are studied by single factor test and orthogonal test to obtain the optimum conditions of extracting polysaccharide in Ligusticum chuanxiong Hort by enzyme.The optimum conditions of extracting polysaccharide in Ligusticum chuanxiong Hort.determined by single factor test,are as follows: cellulose enzyme 0.15%,compound pectic enzyme 6%,pH 3.5,reaction time 150 min and reaction temperature 60 ℃.On the basis of the single factor test,orthogonal test is carried out and the results show that the optimum conditions,on which the extraction rate of polysaccharide is 3.03%,are cellulose enzyme 0.15%,compound pectic enzyme 6%,pH 3.5,reaction time 210 min and reaction temperature 60 ℃.It is simple and fast to extract polysaccharide in Ligusticum chuanxiong Hort by enzyme.And the exaction rate is also higher.
