摘  要:目的:从山药皮中提取总甾体皂甙元并测定其含量。方法:采用超声萃取技术提取山药皮中的总甾体皂甙元。以薯蓣皂甙元为对照品,采用分光光度法测定总甾体皂甙元含量。结果:在单因素试验基础上做L9(34)正交试验,确定最佳提取条件为:超声15min,5%硫酸溶液中水解2h,石油醚回流萃取4h,在此条件下,总甾体皂甙元的提取率为1.467%,相对标准偏差(RSD)为0.89%(n=3)。结论:本测定方法简便、快速,稳定性及重现性好。运用超声处理技术从山药皮中提取总甾体皂甙元效果较好。Objective: To extract and determination total steroidal sapogenins in Chinese yam skin. Methods: Total steroidal sapogenins in Chinese yam skin were extracted by ultrasonic extraction technique and determined its content by ultraviolet spectrophotometry using diosgenin as standard sample. Results: The optimal extraction condition was studied through orthogonal tests [(L9(3^4)] on the basis of single factor test. The extraction rate of total steroidal sapogenins was 1.467% in the optimal extraction conditions were as follows: ultrasonic treatment time 15 min, concentration of sulfuric acid 5%, hydrolysis time in the sulfuric acid 2 h and extraction time in petroleum ether 4 h and RSD was 0.89%(n=3). Conclusion: The determination method was easy, rapid and with good stability and repeatabitity. The results are satisfactory to extract total steroidal sapogenins in Chinese yam skin using ultrasonic technique.
