摘 要:利用以壳聚糖为唯一碳源的培养基从土壤中分离出两株具有产壳聚糖酶能力的菌株,采用平板分离初筛和摇瓶培养复筛,确定了菌种Y4为产壳聚糖酶较好的菌株.通过对菌株Y4生理生化试验、菌株形态特征和生物学特性研究,发现菌株Y4与《伯杰细菌鉴定手册》(第8版)上施氏假单胞菌(Pseudomonas stutzeri)相符,可以判断菌株Y4为施氏假单胞菌,同时,对菌株Y4产酶条件进行了研究,结果发现在pH为6.5,温度为40℃,培养时间为24 h时酶活力达到最大.筛选出来的施氏假单胞菌菌株Y4具有较好的壳聚糖酶活力,为壳聚糖的降解提供了一条新途径.This research mainly included the screening and identification of chitosanase-producing microorganism by method of the see-through circle, strain Y4 for producing chitosanase was screened and chosen. According to the physiological and biochemical experiments and the strain morphological and biological features of the strain Y4, it was tentatively identified that strain Y4 well confirmed to the Pseudomonas stutzeri by Bergeys Manual of Determinative Bacteriology (the eighth edition). Therefore it can be considered that Y4 is Pseudomonas stutzeri Y4. Meanwhile, its enzyme-producing conditions were investigated. The results showed the enzyme activity achieved the maximam when it was cultured for 24 hours at the condition of pH value 6.5 and the temperature 40 ℃. The screen of this research comes out that Pseudomonas stutzeri Y4 have certain enzyme activity. It offered a new route to degrade chitosan into chitosan oligosaccharides that have physiological reactivity.
