摘 要:采用紫外线和亚硝酸对出发菌株001进行诱变和筛选各2轮,获得变异菌株GS228,多糖产量提高了1倍,而且无色素产生或色素淡化。通过比较菌落特征,变异菌株GS228菌落大小、质地和颜色明显优于出发菌株001。对诱变菌株GS228进行遗传稳定性实验,连续培养10代,结果表明:发酵液最终颜色为白色,最终pH为3.2-3.4,发酵液吸光值为0.280-0.295,多糖产量为20.11-21.09g/L,表明变异菌株GS228是稳定的,可应用于实际生产中。The ultraviolet radiation and nitrite were used to treat parental pullulans strain 001 two times,and then a mutant strain GS228,which was pullulanase yield higher one time than 001 and pigment-free or reduced pigments, was obtained. Compared the colony characteristicses between GS228 and 001,the colony size,texture and color of GS228 were obviously better than 001. Genetic stability test of GS228 that was subcultured l0 generations: fermentation fluid final color white, pH 3.2-3.4, absorbency 0.280-0.295 and polysaccharide yield 20.11-21.09g/L, indicated that mutant strain GS228 was stable,may apply in the actual production.
