摘  要:建立了改进的海参体壁组织的扫描电镜(scanning electron microscope,SEM)和透射电镜(transmission electron microscope,TEM)制样方法。SEM样品采用戊二醛固定、乙醇梯度脱水、CO2临界点干燥等处理,与直接冷冻干燥制样法相比,可以最大限度上避免因前处理导致的海参组织变化并消除表面张力的影响;TEM样品采用了先用盐酸胍预处理和人造海水(artificial sea water,ASW)平衡后再进行样品固定的方法,与对样品直接固定的常规方法相比,更有利于保持海参体壁胶原纤维的横纹结构。结果表明:改进的SEM和TEM制样方法能较好地保持样品的微观结构,有利于研究海参体壁组织在加工过程中的超微结构变化。In this study, the improved SEM and TEM sample preparation procedures for sea cucumber body wall were established. To replace the conventionally direct freez.e - drying method, the sample was fixed in glutaraldehyde, dehydrated with series of ethanol, and dried by supercritieal CO2 in SEM sample preparation. Experimental results indicated the new procedure could avoid the artificial alterations in sea cucumber tissue caused by pretreatment and abolish the impact of surface tensile strength. In TEM sample preparation, the specimens were pretreated with guanidine HCl and then equilibrated with artificial sea water before fixation, it conduceed to maintain the integrity of transverse striation structure of collagen fibers compared with the routine procedure without these pretreatment. The results indicated that the improved measures taken here for SEM and TEM sample preparation facilitate the maintenance of the microstructure of sample, and consequently favored the study of changes in microstructure of body wall during processlng.
