摘 要:针对嗜酸乳杆菌的营养需要,采用单因素试验确定MRS培养基中各物质的增菌效果,以及五种生长因子的增菌效果,然后选出四种对嗜酸乳杆菌增菌效果明显的物质,正交试验优化四种物质的用量,得出最佳培养基配比:酵母膏1.5%,牛肉膏1.5%,西红柿汁15%,黄瓜汁20%,以最优增菌培养基测定生长曲线、pH值和滴定酸度的变化,确定增菌培养终止时间为14 h,此时活菌数达1.2×1012 cfu/mL。According to the nutritional needs of LactobaciUus acidophilus Single-factor test to determine enriching effect of substances of the MRS medium, as well as enriching effects of five growthpromoting factors, and then selected four of clear enriching effects material, basis culture medium and growth promoting factors were optimized by orthoganal test. The optimum culture medium composition was as following: 1.5% yeast extract, 1.5% beef extract, 15% tomato juice, 20% cucumber juice, Growth curve with optimized stimulated culture was determined by following the change of pH and titration acidity. According to the growth cure, the terminal time was in 14 hours and Lactobacillus acidophilus got to 1.2 × 10^12 cfu/mL.
