摘 要:对Candida shehatae发酵稻草纸浆水解液生产乙醇的工艺进行了初步研究。分别对影响酶水解和发酵阶段的各因素进行了优化。结果表明,利用正交试验确定的最佳酶水解条件为:酶用量150U/g料,底物浓度2%,反应温度55℃,pH4.8,反应时间8h,酶解得率可达到73.20%。初始葡萄糖浓度为60g/L,装液量100mL/250mL,无机氮源为(NH)42SO4,转速为100r/min,温度为28℃,pH值为5.5,发酵时间为48h,在此条件下乙醇得率可达49.72%,能达到理论得率的97%,转化率最高为0.36g/g(乙醇/稻草纸浆)。Ethanol production from straw paper pulp hydrolysated by Candida shehatae was investigated in this paper. The conditions that impact the enzyme hydrolysis and fermentation were optimized, respectively. The optimal hydrolysis conditions were obtained through orthogonal test as followed: enzyme concentration 150 U/g material, substrate concentration 2%, temperature 55℃, pH 4.8 and hydrolysis time 20 h. Under these condtions, the yield of reducing sugar hydrolysis is 73.2% ;The optimal conditions of ethanol fermentation were as followed: initial glucose concentration 60 g/L, volumes of shaking flask 100 ml/250 ml, inorganic nitrogen source (NH4)2SO4, rotation speed 100 r/min, pH 5.5, fermentation time 48 h. Under these conditions, the ethanol yield rate reached 49.72%, which was 97.0% of the ethanol theoretical yield. The high conversion was 0.36 g/g (ethanol/srraw paper pulp).
