摘 要:以活性碳为载体,采用吸附培养的方法将发酵性丝孢酵母细胞固定化,确定了吸附培养固定化的适宜条件,并测定了固定化细胞的保存稳定性和使用稳定性。结果表明,在100mL发酵培养基中(250mL摇瓶)添加1.0g活性碳于30℃、160r/min吸附培养48h,可吸附较多细胞。将固定化细胞于4℃保存30d脂肪酶活力无明显下降。用固定化细胞催化大豆油转酯化反应,连续催化5批后酯化率仍超过50%。Trichosporon fermentans cells with lipase were immobilized on active carbon by adsorption. The optimum immobilization conditions and the preservation stability and utilization stability of immobilized cells were determined. The results showed that high absorbtion efficiency was obtained in the following conditions: Trichosporon fermentans cells were adsorbed with 1.0 g active carbon for 48 h by 100ml submerged fermentation in 250 ml shaking flask at 30℃ and 160 r/min. The lipase activity of the immobilized cells kept stable at 4℃ within for 30 d. The esterification rate maintained over 50% after 5 successive batches of transesterification reaction with soybean oil.
