摘 要:目的探讨长期高脂高能量饮食情况下膳食脂肪酸对子鼠脑组织脂肪酸组成及脂肪酸合成相关基因表达的影响。方法8周龄健康雌性C57BL/6J小鼠,适应性喂养1周后,雌雄鼠以2∶1比例合笼,检查阴栓确定怀孕后进入实验。按体重将24只雌鼠随机分为对照组、亚麻油组和猪油组,分别饲以基础饲料、亚麻油饲料和猪油饲料。子鼠断乳后,仍继续给予同母代的饲料喂养至第9周。脱臼处死后取子鼠脑组织,以气相色谱法检测其脑组织中不同脂肪酸水平,逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)法检测脑组织β-actin、lxra、acc、fas和scd-1基因的表达。结果与对照组相比,猪油组饱和脂肪酸的水平显著增加(P<0.01),而多不饱和脂肪酸的水平尤其是DHA水平显著降低(P<0.05);与亚麻油组相比,猪油组饱和脂肪酸水平高于亚麻油组(P<0.01),多不饱和脂肪酸的水平尤其是DHA水平低于亚麻油组(P<0.01);与对照组相比,亚麻油组的多不饱和脂肪酸的水平略有增加,但差异无统计学意义。与对照组相比,亚麻油组子鼠脑组织lxra、acc和scd-1基因表达明显上调(P<0.01),而猪油组子鼠脑组织的fas基因表达明显上调(P<0.05)。结论长期高脂高能量饮食情况下膳食脂肪酸可以影响子鼠脑组织脂肪酸组成,其机制可能与调控脂肪酸合成相关基因的表达有关。Objective To observe the long-term impact of dietary fatty acids on fatty acids composition and relative gene expression of brain in the offspring of C57BL/6J mice fed with high fat high energy diet. Method Eight-week-old healthy C57BL/6J mice, after adaption for one week, were caged with a ratio of 2: 1 for female and male. Mating was confirmed in the next morning by the presence of vaginal plug. Twenty four dams were housed individually and divided into three groups fed with normal chow diet, linoleum oil diet or lard diet respectively. The offspring were fed with the same diet with their mother from ablactating until 9 weeks old. Fatty acids of brain were analyzed by gas chromatography and expressed as percent (%) of total fatty acids. The relative expression of β-actin, lxra, acc, fas and scd-1 genes were analyzed by RTPCR. Results The composition of SFA (saturated fatty acid ) were significantly increased, whereas PUFA ( polyunsaturated fatty acid) , especially DHA ( docosahexaenoic acid) , was significantly decreased in the offspring of lard diet group compared with that of normal chow diet group. The expression of lxra, acc and scd-1 were up-regulated in the offspring of linoleum oil diet group, and the expression offas was up-regulated in lard diet group. Conclusion The brain fatty acid composition of the offspring from the dam fed with high fat high energy diet might be affected by regulating the expression of relative gene of fatty acid synthesis.
