摘 要:研究了树干毕赤酵母CBS5773在高浓度木糖条件下发酵乙醇的情况,结果表明,在木糖浓度为200g/L时,树干毕赤酵母仍具有较高产乙醇能力,达0.48mL/10mL。单一添加硫酸铵作为氮源,考察不同pH值、温度、溶解氧对产乙醇的影响,发酵条件为pH4.5、25℃、130r/min时效果最佳。添加不同氮源试验结果表明,添加尿素比添加硫酸铵的效果高出25%,最佳发酵条件为木糖200g/L,蛋白胨0.5g/L,酵母浸膏1.0g/L,CaCl20.25g/L,MgSO40.25g/L,KH2PO42.5g/L,尿素1.0g/L,pH值为4.5,温度30℃,速率150r/min。Ethanol fermentation using Pichia stipitis CBS5773 in high concentration of xylose were conducted by different fermentation conditions, substrate concentration. The fermentation capability of the P. stipitis CBS5773 in high concentration of substrate concentration was investigated, and the studies showed that P. stipitis CBS5773 still had a higher capacity of ethanol fermentation, which was up to 0.48 ml/10 ml. Adding single (NH4)2SO4 as nitrogen source, the effect of P. stipitis CBS5773 fermentation on different pH, temperature, dissolved oxygen were analyzed, and the results showed that the optimal fermentation conditions is pH 4.5, 25℃, 130 r/min. In the high substrate concentration, adding different nitrogen sources to ferment, the results showed that adding urea can get 25% higher ethanol yields than adding (NH4)2SO4. The best conditions for xylose fermentation is 200g/L, peptone 0.5g/L, yeast extract 1.0g/L, CaCl2 0.25g/L, MgSO4 0.25g/L, KH2PO4 2.5g/L, urea 1.0g/L, pH 4.5, 30℃, 150r/min.
