摘 要:研究筛选产淀粉酶海洋菌并对发酵条件进行了优化。采用3,5-二硝基水杨酸试剂显色法对菌种复筛;研究了碳源、氮源、各种理化因素等培养条件对细菌产淀粉酶的影响。确定W11菌为研究菌,初步鉴定为弧菌;最佳碳源为麸皮,最佳氮源为蛋白胨,最适初始pH值为7.04~7.70。培养温度、接种量、装液量对细菌产酶有较大影响,正交试验表明:麸皮10g/L,蛋白胨10g/L,250mL三角瓶中装液75mL,温度33℃时,酶活可达到325.14U/mL。比未优化前提高了2.71倍。Screening of marine bacterium and optimization of fermentation conditions for amylase production were studied. The strain was finally de- termined with chtomogenic method with 3,5-dinitrosalicylic acid and effects of carbon source, nitrogen source, and physicochemical factors on pro- duction of amylase by bacterium were also studied. Strain Wl 1 was used for further study and it was preliminarily identified as Vibrionaceae. Opti- mum sources of carbon and nitrogen were wheat bran and peptone, respectively with initial pH at 7.04-7.70. With orthogonal test, the optimum con- ditions were determined as bran 10 g/l, peptone 10 g/l, with 75 ml medium in 250 ml shaking flask and inoculated under 33℃. Under the optimum conditions, enzyme activity could reach 325.14 U/ml, which was 2.71 times as normal conditions.
