摘 要:以挤压膨化豆渣为原料,采用纤维素酶酶法制备高活性膳食纤维。通过单因素试验及L9(34)正交试验对酶解条件进行优化。试验结果表明,纤维素酶最适作用条件为:pH5.0,温度50℃,加酶量2%,反应时间2.5h,料水比1∶18(m∶v)。在此条件下,豆渣中水溶性膳食纤维(SDF)得率为23.9%。采用高效凝胶过滤法测定SDF的分子量分布发现,经挤压酶解处理后的豆渣的SDF的组分发生了变化。Extruded soybean dregs was used as raw material to prepare for soluble dietary fiber by enzymatic hydrolysis. The effects of the ratio of solid to liquid, temperature, extraction time and enzyme concentration and pH value on the yield of soluble dietary fiber were studied respectively. Based on the results, the orthogonal experiment was designed to obtain optimized technology for preparing soluble dietary fiber. The results showed that the optimum enzymatic hydrolysis conditions were 50 ℃, pH 5.0, enzyme concentration 2%, 2.5 h, ratio of substrate to water 1∶18. Under optimal conditions,the yield of soluble dietary fiber was increased to 23.9%. The molecular weight distribution of SDF was analyzed by high performance gel filtration chromatography, and the results showed that the SDF composition of soybean dregs treated by double screw extruding was changed.
