摘 要:为了制备高活性醒酒玉米肽,采用了超滤技术对酶解液进行分级分离,通过体内外试验比较了各级分的醒酒玉米肽,并研究了pH对玉米肽超滤过程的影响。结果显示:体外试验中活性较高的分子质量小于5 ku的玉米肽级分,在动物试验中表现出最高的血醇清除率,达34.35%;与原酶解液相比,活性极显著提高(P〈0.01)。随着时间的增加,在pH 6、7、8、9条件下,超滤膜通量和传质率(Tr)不断下降,而浓缩体积倍数(VCF)不断增加;而同一时间,随着pH的增加,膜通量和VCF逐渐增加,而Tr则逐渐下降。采用截留分子质量为5 ku的超滤膜对玉米肽进行分级分离,膜透过物(即Mw〈5 ku)的醒酒活性可极显著提高(P〈0.01);碱性条件下(pH 8~9)对玉米蛋白酶解液进行超滤效果最好。Enzyme hydrolysis combined with ultrafiltration technology were used to obtained corn peptides(CP) with high sober-up activity from corn protein;the sober-up activities of CP fractions were compared with in vitro test and animal test;additionally,the effect of pH on the ultrafiltration was evaluated.Results: CP fraction of molecular weight(Mw) 5 ku which displays relatively higher sober-up activity in vitro,gains a maximum eliminating rate of blood alcohol concentration(34.35%) in mice,an extra significant(P0.01) enhancement of eliminating blood alcohol concentration compared with original hydrolysate.The transmission(Tr) and flux decrease,while volume concentrated factors(VCF) increase with time extending during the ultrafiltration at all test pH.However,the flux and VCF increase,while Tr decreases,with the increase of pH at the same time.Conclusion:The sober-up activity of CP fraction of Mw 5 ku is significantly enhanced after fractionation using ultrafiltration membrane with 5 ku molecular cut off,compared with original hydrolysate(P0.01).Adjusting pH value at 8~9 would be the best choice during the ultrafiltration of corn protein hydrolysate.
