摘  要:建立了活性电泳分析蛋白酶组分的方法。在制备分离胶时,加入终浓度为0.5%的SDS和1%的酪蛋白,4℃下电泳,以2.5%Triton-X100洗去SDS,在不同条件下温浴,可得到清晰的蛋白酶谱。采用该方法在沪酿3.042的成曲浸提液中检测到7种蛋白酶组分,这些组分在pH中性条件下都有活性,其中3种蛋白酶在pH酸性条件下仍然有很强的活性。这些发现与对米曲霉蛋白酶的传统认识有很大的不同。A method for analysis of protease components of active electrophoresis was established. Clear zymography could be observed by the proce- dures as follows: 0.5% SDS and 1% casein was added in resolving gel, running electrophoresis was carried out under 4℃, removing SDS with 2.5% Triton-X100 and culturing the gel in different pH buffer. Seven protease components could be detected from Aspergillus oryzae Hu-niang 3.042 by this method. All the seven components were active under neutral pH value, and three of them have high activity under acidic pH value, as well. The results differed from the traditional knowledge ofAspergillus oryzae Hu-niang 3.042 proteases.
