摘　　要：研究乳酸菌亚硝酸盐还原酶制备。将细胞破碎、硫酸铵盐析从乳酸菌中提取亚硝酸盐还原酶,经DEAE-52纤维素柱层析、Sephadex G-150凝胶过滤纯化,获得酶制品经SDS-聚丙烯酰胺凝胶电泳法测得相对分子质量约为42kDa。酶学性质分析表明,最适温度30℃,最适pH 5.5,以亚硝酸钠为底物,酶的Km值为120.5ug/mL。The nitrite reductase was purified by ammonium sulfate precipitation, DEAE-52 cellulose column chromatography, Sephadex G-150 gel filtration. Higher purity ofreductase was prepared. Molecular weight of the nitrite reductase subunit was about 42kDa by SDS-PAGE. The properties of purified enzyme showed The optimal temperature and pH of enzyme reaction were 30℃ and pH 5.5 respectively. The apparent Km value for nitrite reductase was 120.5ug/ml with sodium nitrite as the substrate.
