摘  要:用反相高效液相色谱法(RP-HPLC)测定不同品牌啤酒中α-酸的总含量,以Kromasil C18(150mm×4.6mm ID,5μm)为色谱柱,以甲醇∶水(85∶15,v/v,85%磷酸调pH值为2.5)为流动相,流速1.0mL/min,紫外检测波长为313nm,外标法定量,α-酸的线性范围为0.08mg/mL^0.80mg/mL,加标回收率在96.66%~101.83%之间,RSD为1.66%,最低检测浓度均为0.001mg/mL。方法具有简单、快速、准确等特点。Contents of α-acids of 8 kinds of beer were determined with reversed-phase high performance liquid chromatography (RP-HPLC). The optimum conditions were: column Kromasil C18 (150mm×4.6 mm ID, 5 μm), mobile phase ratios of methanol to H20 by 85:15 (v/v) and adjust pH value to 2.5 with phosphoric acid, flow rate was 1.0mg/min, detected with ultraviolet detection at wavelength of 313nm. A good linear relationship of α-acids was found in the range of 0.08mg/ml-0.80mg/ml. The average recovery of α-acids was 96.66%-101.83%, and relative standard deviation (RSD) of the method was 1.66%. The method is simple, rapid and has high accuracy.
