摘  要:从8株固态发酵高产红曲色素菌株中筛选出一株适合于液相培养工艺的高产红曲色素菌株FL1,并对其液相培养过程特性及高产色素低产桔霉素的代谢调控策略进行了研究。发酵过程动力学研究结果表明,红曲色素与桔霉素作为次级代谢产物,其合成与红曲霉菌生长的关系属于非生长偶联型。本文首次采用连续补料的Fed-Batch培养技术,用于红曲霉菌的液相培养。在初期菌体生长阶段,红曲霉菌的最大比生长速率μ_m达到0.21h^(-1);中后期通过连续补加糖液,可将还原糖浓度控制在5~10g/L,从而将μ控制在合适范围(0.02h^(-1))。补料培养工艺有效的延长了红曲霉菌合成色素的时间,大大提高了色素产量。红曲霉菌的Fed-Batch培养过程可控性强,重复性良好,平均色素得率和时空得率高达5400U/g淀粉和5625U/(h·L),显著高于目前国内外已报道的红曲霉菌液相发酵水平。FL1, the high Monascus pigment producing strain for liquid-phase culture was screened form 8 high Monascus pigment producing strains in solid-phase culture. The liquid-phase culture characteristics and metabolic regulation strategy for high-yielding pigment and low-yielding citrinin of FL1 were studied. Results of kinetics studies showed that the Monascus pigment and citrinin, as. secondary metabolites, the relationship between their synthesis and the growth of Monascus was non-growth-coupled. The fed-batch culture technology was first used as a new method in liquid-phase culture. The biomass specific growth rate of Monascus achieved 0.21 h^-1 in growth phase; in mid and late period, reducing sugar concentration could be controlled at 5 - 10 g/L by continuous feed. Thereby, the biomass specific growth rate of Monascus could be controled within a suitable range(0.02 h^-1). The fed-batch technology enhanced pigment accumulation time and greatly increased the pigment production. This new culture technology had a good reproducibility and controllability. The average Monascus pigment yield rate and space time yield rate could be achieved 5 400 U/g starch and 5 625 U/(h·L), obviously more than those reported at home and abroad.
