摘 要:为优化磨盘柿中多酚类物质的提取和纯化工艺,在前期单因素试验的基础上,采用正交试验,以Folin-Ciocalteu比色法为总多酚提取率的测定方法,探讨乙醇体积分数、提取温度、时间、液固比4个因素的影响,确定最佳提取参数。根据对没食子酸的静态吸附及解吸性能,从9种树脂中筛选出具有最佳分离纯化效果的一种树脂,并研究其对柿子多酚的静态及动态吸附、解吸效果。研究结果表明,当乙醇体积分数60%,温度55℃,提取时间30min,液固比(mL:g)15时多酚提取率最高,3.0667mg/g鲜柿;HP2MGL大孔树脂对柿子多酚的分离纯化效果最好,静态吸附容量达到25.44mg/g,60%的乙醇洗脱剂解吸效果最好,解吸率为63.09%;动态试验表明,树脂对柿子多酚的吸附速度较快,不同浓度乙醇可有效洗脱出不同的极性组分。To obtain the optimum extraction and purification technology of polyphenols from Mopan persimmon,the conditions of extraction were optimized by orthogonal experiment on the basis of prior single-factor experiments.The extraction of total polyphenols from persimmon was determined by Folin-Ciocalteu Colorimetry.HP2MGL was selected from 9 different types of macroporous resins and the static and dynamic properties of adsorption and desorption have been investigated.Results indicated that the optimum conditions of extraction were as follows:60% ethanol solution,ratio of liquid to solid 15:1(mL/g),55 ℃,30 min.In this condition,the highest extraction rate of persimmon polyphenols was 3.0667 mg/g FW.HP2MGL macroporous resin had perfect effect on separation and purification of persimmon polyphenols.Static adsorption capacity was 25.05 mg/g,desorption rate of 60% ethanol solution was up to 65.47%;Dynamic experiments showed that the adsorption speed of HP2MGL was fast,and ethanol solution of different concentration could effectively elute different polarity compositions of polyphenols.
