PEF协同酶提取猪皮蛋白工艺条件的研究

摘  要:研究高压脉冲电场(PEF)对枯草杆菌中性蛋白酶提取猪皮胶原蛋白的影响及提取的工艺条件。首先考查高压脉冲电场强度、脉冲数、酶用量、酶解时间等因素对生猪皮胶原蛋白提取效果的影响,然后以此为依据采用正交法优化提取条件。试验结果为,经高压脉冲电场处理的猪皮,其胶原蛋白的溶出率远高于未经高压脉冲电场处理的猪皮,而且酶解所用的时间短。在料液比1∶10(g/mL)、温度40℃、pH=6.5、酶用量10.0μkat/g、电场强度25 kV/cm、脉冲数40次、酶提时间30 min等条件下,胶原蛋白溶出率可达84.2%。The effect of PEF on the yield of extracting collagen protein from pig skin by bacillus subtilis neutral proteinase and the extracting technological conditions was researched in this paper. The experimental processes were that the influence factors on extracting yield those were electric intensity, pulse number, enzymes usage and enzymolysis time were studied at first, and based on it, the technological conditions of extraction were defined by orthogonal test following. The results of the experiment showed that the dissolution ratio of the extracting method preprocessed by PEF could be more higher than the un-preprocessed one,and it could save more enzymolysis time for this method. Under the conditions of material to liquid ratio 1:10 (g/mL), temperature 40℃, pH=6.5, enzymes usage 10.0 μkat/g, electric field intensity 25 kV/cm, pulse number 40 and enzymolysis time 30 min, the dissolution ratio could reach to 84.2 %.
