摘 要:探讨了培养基3种初始pH对木聚糖酶合成的影响。结果表明,培养基初始pH对木聚糖酶的合成有重大影响,低pH有利于提高木聚糖酶的活力。在碳源为7 g/L的条件下,初始pH为4.0时,合成的木聚糖酶活力高达32.87 IU/mL,酶产率和得率分别为6574.0 IU/L·d和4 695.7 IU/g木聚糖。酶活力和酶产率均是初始pH为5.0时的1.7倍,分别是初始pH为6.0时的3.8倍和2.3倍。进一步研究表明,初始pH为4.0更有利于提高β-木糖苷酶的活力。Initial pH of medium had important influence on xylanase synthesis by Trichoderma reesei Rut C30. The results showed that low pH was beneficial to increasing the activity of xylanase. Among three initial pH of media to induce xylanase, the optimal initial pH was 4.0, and initial pH 6.0 was poor. When carbon source was 7 g/L and initial pH was 4.0,the activity of xylanase was up to 32.87 IU/mL, with the productivity and yield being 6 574.0 IU/(L·d) and 4 695.71 IU/g xylan respectively. Further study indicated that initial pH 4.0 of medium was more beneficial to increasing β-xylosidase activity. This means the influence of pH on the synthesis of β-xylosidase was greater than that on the synthesis of endo-xylanase.
