摘  要:目的:探讨叶酸-壳聚糖siRNA纳米复合物输送抗肿瘤细胞乙酰肝素酶的siRNA的效率和对乙酰肝素酶基因表达和肿瘤细胞侵袭力的影响。以期提高siRNA的输送效率,实现安全有效的靶向性基因治疗,并为研发抗肿瘤侵袭转移药物提供理论基础。方法:制备FA-CS-siRNA纳米复合物并测定其特征;分别以control组、FA-CS组、FA-CS-siRNA纳米复合物组、CS-siRNA组和lipo2000-siRNA组转染乳腺癌MCF-7细胞,RT-PCR和Western Blot检测乙酰肝素酶表达情况;肿瘤侵袭实验检测MCF-7细胞侵袭转移能力的变化。结果:红外光谱确认叶酸已成功与壳聚糖偶联;FA-CS-siRNA纳米复合物形态为圆形,分布较均匀,平均粒径为197.7nm;FA-CS-siRNA纳米复合物组能有效地干扰MCF-7细胞中乙酰肝素酶的表达;肿瘤细胞侵袭实验显示lipo2000-siRNA组和FA-CS-siRNA纳米复合物组能有效抑制MCF-7细胞的侵袭和转移。结论:FA-CS-siRNA纳米复合物能有效传递siRNA进入MCF-7细胞。Objective:To investigate the delivery efficiency of heparanase-specific siRNA which were loaded by folate-chitosan-siRNA nanocomposites and the effect of heparanase-specific siRNA on heparanase gene expression and tumor cell invasion and metastasis in vitro,so as to improve the efficiency of siRNA delivery and to achieve safe and effective targeting gene therapy and to provide a theoretical basis for the exploiting of the anti-tumor drug. Methods:Prepare the folate-chitosan-siRNA nanocomposites and determine its characteristics. The transfection assay was divided into 5 groups:control group,FA-CS group,FA-CS-siRNA nanocomposites group,CS-siRNA nanocomposites group and Lipo2000-siRNA group. Then the heparanase expression was detected by RT-PCR and Western Blot,tumor invasion assay suggested the change of MCF-7 cell invasion and metastasis. Results:The FA-CS was verified by the infrared spectroscopy. The FA-CS-siRNA nanoparticle was round-form,distribute evenly,and the particle size was 197. 7nm. FA-CS-siRNA nanocomposite group could interfere with heparanase expression effectively in MCF-7 cells,tumor cell invasion assay showed that lipo2000-siRNA group and the FA-CS-siRNA nanocomposites group could inhibit the MCF-7 cell invasion and metastasis. Conclusion:FA-CS-siRNA nanocomposite could effectively transfer siRNA into MCF-7 cells.
