双菌比较优化稻草“液化”发酵产乙醇的研究

摘  要:利用稻草液化产物为底物,分别采用酿酒酵母和休哈塔假丝酵母发酵生产乙醇,对影响发酵阶段的各因素进行优化,选取最佳菌种完成秸秆到乙醇的转化。结果表明,液化产物经酶解后葡萄糖浓度可达69.5mg/mL,是发酵制备乙醇的良好底物。优化发酵后,酿酒酵母更适合做液化产物的发酵菌种。适宜的发酵条件:初始葡萄糖浓度60-65mg/mL,温度30℃,pH=6.0,装液量80mL,接种量10%,发酵时间36h,在此条件下乙醇得率可达49.3%,能达到理论得率的96.1%,转化率最高为0.27g,g(乙醇/液化产物)。With the use of product from liquefied straw as the substrate, ethanol was prepared by fermentation of Saccharomyces cerevisiae and Candida shehatae respectively. The effect factors of fermentation stage were optimized in order to select the optimal strain to complete the transformation of straw into ethanol.The results showed that the glucose density might reach 69.5 mg/mL after the enzymolysis of liquefied product,which was a good substrate for ethanol fermentation preparation. Saccharomyces cerevisiae was more suitable for fermentation microbial strain after the optimized fermentation condition.Under the suitable fermentation conditions: initial glucose density 60- 65 mg/mL, the temperature 30 ℃, pH 6.0,with fluid amount 80 mL triangle bottle, inoculation amount of 10%, fermentation time of 36 h, the ethanol yield rate might reach 49.3%, which achieved the theory yield rate 96.1%. The highest conversion rate was 0.27 g/g(ethanol/straw liquefied products).
