摘 要:通过采用10%三氯乙酸水溶液对老陈醋样品进行预处理,以邻苯二甲醛(OPA)、氯甲酸-9-芴基甲酯(FMOC-CL)为衍生剂,用反相C18柱为分离柱,柱温40℃,梯度洗脱,在EX:250nm,EM:410nm下检测,建立了OPA柱前自动衍生-荧光检测老陈醋中γ-氨基丁酸(GABA)的HPLC法。结果表明,老陈醋中的GABA获得了很好的分离,GABA在0.004~0.30mg/mL范围内线性相关性好,其线性方程为Y=26016X-38.72(R2=0.9994),加样平均回收率达到了96.22%以上。该方法稳定、灵敏、重现性好,可用于测定老陈醋中GABA含量定量分析。A method for γ-Amino Butyric Acid(GABA) determination in mature vinegar with ortho-phthaladehyde(OPA) automatic pre-column derivation and fluorescence detection by high performance-liquid chromatography was established.Firstly mature vinegar was pretreated with trichloroacetic acid of 10%,derived OPA and 9-fluorenylmethyl chloroformate(FMOC-CL),and then separated and assayed with reversed phase C18 column,finally eluted at 40℃ and determinated by HPLC at EX:250nm,EM:410nm.The result showed that GABA in mature vinegar was separated well with this method,and in the range of 0.004~0.30mg/mL of calibration curve,GABA kept linear with regression equation of Y=26016X-38.72(R2=0.9994).The relative error was 96.22%,which indicated that method was a sensitive,reproduced and specific assay for the determination of GABA in mature vinegar.
