摘 要:目的:研究乌梅提取物脱色工艺及抑菌机理,为乌梅的高效利用以及新型食品生物抑菌剂的开发提供理论基础。方法:选用活性炭、AB-8大孔树脂、中性氧化铝对乌梅提取液进行脱色;采用杯碟法、倍半稀释法分别测定抑菌圈和最低抑菌浓度;通过扫描电镜观察乌梅提取物对受试菌的影响,对抑菌机理作初步分析。结果:对脱色工艺的比较研究表明,选用AB-8大孔树脂,在工作流速为2~3BV/h,脱色率为98.32%时,脱色后抑菌活性损失率仅5.5%;抑菌试验表明,乌梅提取物脱色后对大肠埃希氏菌、金黄色葡萄球菌、枯草芽孢杆菌及沙门氏菌的抑菌圈直径分别为25.58、25.30、22.52、23.84mm,MIC分别为3.12、3.12、6.25、6.25mg/mL;扫描电镜结果表明,乌梅提取物作用后的大肠埃希氏菌细胞结构被破坏,细胞内容物外泄,而金黄色葡萄球菌形态变化不明显。结论:选用AB-8大孔树脂进行脱色效果最好。乌梅提取物对大肠埃希氏菌的抑制作用是因其对菌体细胞壁和细胞膜的破坏作用,而其对金黄色葡萄球菌的抑制作用有待进一步研究。Objective:The preparation progress and antibacterial activity of Fructus mume extract has researched to provided theoretical foundation to high efficiency utilization and the development of new biological food antibacterial agents.Method:Comparative analysis of the decolorization of Fructus mume extract using activated carbon,macroporous resin AB-8 and neutral alumina.Fructus mume extract were used for antibacterial activity(ABA) testd by oxford cup test,and tested the minimum inhibitory concentrations of the extracts with test tube times dilution.Analyse antimicrobial mechanism of Fructus mume extract by observe the tested bacteria through the scanning electron microscope.Results:Comparative analysis showed that the decolorization efficiency of macroporous resin AB-8 with its velocity of flow 2~3BV/h was much better than others,the decolorizating rate come to 98.32% when the antibacterial activity material reserved rate was 94.5%.It also evident that Fructus mume extract decoloring by macroporous resin AB-8 has antibacterial activity to Escherichia coli,Staphyloccocus aureus,Bacillus subtilis and Salmonella,the inhibition zone diameters was 25.58,25.30,22.52,23.84 mm and the MIC(Minimum Inhibitory concentration) was 3.12,3.12,6.25 and 6.25 mg/mL.Observation under scaning electron microscope revealed that structure of Escherichia coli cells were significantly destroyed by Fructus mume extract,and the cell wall were melted and the morphology of Staphyloccocus aureus almost no change.Conclusion:The effect of the decoloring with macroporous resin AB-8 is better than the other two kind of good.The inhibition of Escherichia coli is due to the damage of the cell wall and cell membrane.But the antimicrobial mechanism of Staphyloccocus aureus needs futher research.
