摘 要:以小麦面粉为原料,采用水提醇沉法提取水溶性阿拉伯木聚糖(WE-AX),比较了硅藻土吸附法和酶法两种方法在WE-AX提取过程中的蛋白脱除效果.结果表明:硅藻土吸附法操作简便、快捷,但蛋白脱除效率偏低;酶法蛋白脱除效率较高,但操作时间长,能耗高,后续处理困难.采用正交试验确定了酶法脱除蛋白的最佳条件为:中性蛋白酶添加量为0.06%(W/V),反应温度为50℃,反应时间为4 h,蛋白质脱除率为76.20%.同时,结合WE-AX整体提取过程中其他环节的蛋白质脱除情况,可使蛋白质脱除率达到99.7%.In this paper, we extracted water-soluble arabinoxylans (WE-AX) from wheat flour by water extraction and ethanol precipitation method, and compared the protein removal effects of diatomite adsorption method and enzyme method during extraction of WE-AX. The results showed that the diatomite adsorption method was simple and fast, but had low protein removal rate; and enzyme method had high protein removal rate, but long operation time, high energy consumption and difficult post-treatment. The optimum protein removal conditions of enzyme method were determined by orthogonal experiments as follows: neutral protease amount 0.06% (W/V), reaction temperature 50 ~C, and reaction time 4 hours. Under the optimum conditions, the protein removal rate was 76.20%. Since the protein could be removed in other links of the extraction process, the total protein removal rate could reach 99.7%.
