摘 要:以肽得率为指标,应用响应面法对贻贝肉酶解条件进行优化,根据二次正交旋转组合试验设计的原理,选取酶(E)与底物(S)的质量比、酶解温度、酶解时间三因素进行组合试验,建立肽得率的二次回归方程。通过响应面分析得到优化组合条件:酶(E)与底物(S)的质量比3.5∶100,温度56℃,时间4.2 h。以此条件进行试验验证,得酶解液中肽含量为35.04%(质量分数),与响应面模型所预测的肽得率(36.27%)接近,说明该优化方法可行。Taking peptide recovery ratio as target index, response surface methodology (RSM) was applied to optimize the conditions of enzymatic hydrolysis for Mytilus edulis. According to the principle of quadratic orthogonal rotating combinatorial design, enzyme (E) and substrate (S) mass ratio, enzymatic hydrolysis temperature and enzymatic hydrolysis time were chosen as factors to establish a quadfie regression equation for describing the peptide recovery ratio. By response surface analysis, the optimum conditions for enzymatic hydrolysis was obtained as follows, Enzyme (E) and substrate (S) mass ratio was 3.5:100, enzymatic hydrolysis temperature was 56℃ for 4.2 h. The peptide recovery ratio was up to 35.04% under the optimum condition, which was close to the pedicted value of 36.27%. These results demonstrate the validity of this extraction process.
