摘 要:采用响应面法优化钾法提取草鱼加工废弃物中鱼油的工艺,并运用气相色谱对其脂肪酸组成进行分析。在单因素实验的基础上,选择pH、水解时间、水解温度、盐用量作为实验因素,利用Box-Benhnken设计和响应面分析法,通过回归分析确定最佳提取工艺为:pH 9.2,水解时间43min,水解温度58℃,盐用量9.5%,在此工艺条件下鱼油提取率达58.61%。气相色谱分析结果表明,草鱼鱼油中饱和脂肪酸、单不饱和脂肪酸和多不饱和脂肪酸的含量分别为22.63%-26.07%、45.09%-47.93%、25.99%-31.32%。Response surface methodology(RSM) was selected to optimize the extraction process of fish oil from processing waste of grass carp by KOH hydrolysis,and GC was applied to determine the content and composition of fatty acids.Based on the experiments of single factors,pH,hydrolysis time,hydrolysis temperature and the amount of salt were selected for Box-Benhnken central composite designing and RSM.The results showed that the optimum extraction technique parameters were:pH of 9.2,hydrolysis time of 43 min,hydrolysis temperature of 48℃,the amount of salt 9.5%(w/w),under these conditions,the extraction yield of fish oil was up to 58.61%.The percentage of saturated fatty acids(SFA),monounsaturated fatty acids(MUFA) and polyunsaturated fatty acids(PUFA) in fish oil were about 22.63%-26.07%,45.09%-47.93% and 25.99%-31.32% respectively.
