摘  要:目的:研究绿茶成分——儿茶素、咖啡碱和茶氨酸对3T3-L1前脂肪细胞的细胞增殖及脂肪代谢的影响。方法:测定不同浓度儿茶素、咖啡碱和茶氨酸对3T3-L1细胞增殖的影响,确定无毒性浓度。在分化诱导液中添加各绿茶成分后对3T3-L1细胞进行96h诱导分化,分化后第6天测定脂肪细胞中甘油三酯(TG)含量。细胞分化后第9天,单独添加绿茶成分或同时添加去甲肾上腺素(NA)24h,分析对细胞中脂肪分解的影响。结果:添加20μg/mL以上质量浓度的儿茶素能显著抑制3T3-L1细胞增殖,但质量浓度40,80μg/mL的儿茶素对3T3-L1细胞有毒性作用,而160μg/mL咖啡碱和茶氨酸对细胞增殖无明显影响。20μg/mL儿茶素能显著抑制3T3-L1细胞中TG的合成,而咖啡碱和茶氨酸对细胞脂肪沉积无明显影响。与单独添加NA相比,同时添加咖啡碱能显著促进NA诱导细胞中脂肪分解的能力。结论:儿茶素抑制脂肪细胞的增殖和脂肪沉积,咖啡碱促进激素诱导脂肪分解。绿茶成分中儿茶素和咖啡碱对脂肪细胞内的脂肪代谢有调节作用。Objective: To investigate the effects of green tea components, catechins, caffeine and theanine on 3T3-L1 cells proliferation and fat metabolism. Methods: 3T3-L1 cells were cultured in DMEM contained different concentrations of catechins, caffeine or theanine, and analyzed on cells proliferation and cytotoxicity. Then the differentiation of 3T3-L1 cells were induced with these green tea components at noncytotoxic concentration for 96 hours. 6th day after differentiation, triglycerides(TG) in 3T3-L1 cells was measured. Lipolytic effect of green tea components in the present or absent of noradrenaline(NA) for 24 hours in 3T3-L1 cells was analyzed on 9th day after differentiation. Results: Above 20 μg/mL catechins significantly inhibited the proliferation of 3T3-L1 cells, but there was cytotoxic effect 40 and 80 μg/mL. However, caffeine and theanine have no effect to the cell even at 160 μg/mL. 20 μg/mL of catechins remarkably reduced the TG level in adipocyte, but caffeine and theanine have no effect. Catechins, caffeine and theanine alone have no lipolytic effect. However, caffeine significantly improved NA-induced lipolysis in 3T3-L1 cells. Conclusion: Catechins could suppress fat accumulation, while caffeine could improve hormone-induced lipolysis in 3T3-L1 cells. These results suggested that catechins and caffeine have fat metabolism regulating effect.
