摘 要:以多糖提取得率为指标,利用响应面分析法优化乌饭树树叶多糖的提取工艺。先利用单因素试验选取影响因素与水平,在此基础上采用四因素三水平的响应面分析法,利用回归分析优化得到最佳提取工艺:1:45(m:V)的料水比,调pH到9.5,在65℃下提取2.5h,该条件下多糖得率为68.629 1mg/g,多糖纯度为38.27%。进一步优化多糖纯化工艺为提取液与95%乙醇按1:5(V:V)混合,静置6h,多糖纯度达到52.03%。The optimal extraction process of polysaccharides from Vaccinium Bracteatum Thunb. Leaves (VBTL) was established by response surface method (RSM). The optimal extraction process was established by response surface analysis as following: pH 9.5, temperature 65 ℃, duration 2. 5 hour, and material /water ratio 1 : 45(m : V). By this optimized process 68. 629 1 mg polysaccharide could be obtained from one gram VBTL, the purity of VBTL poly saccharides was 38.27%. The orthogonaI experiment was carried out to obtain the optimum conditions of the purification of VBTL poly- saecharides. The optimal conditions were as the following: the alco- hol precipitation static time was 6 h, the volume ratio of extracting agent and alcohol was 1 : 5, the ethanol concentration (V :V) was 95%. By this process, the purity of polysaccharides increased from 38.27% to 52.03%.
