高温产蛋白酶菌株的筛选及产酶条件的研究

摘　　要：从新疆吐鲁番地区土样中分离筛选获得1株高温产蛋白酶菌株,经形态观察和16S rRNA初步鉴定为芽孢杆菌(Bacillus tequi-lensis),将其命名为C9。通过对其产酶条件的研究表明:菌株C9以葡萄糖为最优碳源,硝酸钠为最优氮源,最佳产酶条件为A3B2C1,即在起始pH值为5.0的发酵培养基中,52℃下发酵培养48h时,所产蛋白酶活性最高,其活性为41.653U/mL。One thermophilic bacterial strain with protease activity was obtained from the soil of Turpan of Xinjiang province. Based on morphological characteristics and 16S rRNA gene sequence, strain was identified as Bacillus tequilensis, named C9. The optimum carbon and nitrate source were glucose and sodium nitrate, respectively. The optimum fermentation conditions were as folllowed： initial pH value 5.0, temperature 52℃ and cultiva- tion time 48h. Under the above conditions, the protease activity reached 41.653U/ml.
