摘 要:目前工业生产药用右旋糖酐常用酸解法,能耗高,污染大,而酶解法反应条件温和,反应速度快,可以在右旋糖酐发酵的同时进行,可大大降低能耗,减轻污染。本文通过对右旋糖酐酸水解和酶水解产物的成分和分子量分布的研究,为酶解法生产药用右旋糖酐新工艺打好基础。实验结果表明:酸水解的终产物基本上为葡萄糖,而酶水解终产物为异麦芽寡糖;水解到重均分子量为20000 Da以上时,2种水解方法对右旋糖酐含量和分布系数影响不大;水解到重均分子量小于20000 Da之后,酶解产物右旋糖酐含量下降。Acidic hydrolysis is a common used process for clinical dextran. However, it causes high energy consumption and heavy pollution. Enzymatic hydrolysis has advantages of moderate conditions and fast reaction rate, and it can be carried out to control molecular weight during the process of dextran fermentation. The primary research of this paper concemtrates on the composition and molecular weight distribute of the dextran hydrolyzates of both acidic and enzymatic hydrolysis. The results show that, (1)Most of the end products of acidic hydrolysis are glucose, whereas the end products of enzymatic hydrolysis are isomalto-oligosaccharides; (2)The content and the polydispersity index of the dextran have no difference between acidic and enzymatic hydrolysis end products when the average molecular weight (Mw) is higher than 20000 Da, the content of enzymatic hydrolysis dextran decreases when the Mw is lower than 20000 Da.
