摘  要:以富硒碎米为原料,研究了大米含硒蛋白提取条件、硒代多肽的酶法制备及其抗氧化活性。结果表明:NaOH浓度为0.07 mol/L、料液比1∶20(m/V)、40℃下提取3 h时可获得蛋白含量、硒含量分别为80.18%和0.583 mg/kg的大米含硒蛋白。相对于木瓜蛋白酶和胃蛋白酶,胰蛋白酶水解大米含硒蛋白可获得具有更高抗氧化活性的多肽。在胰蛋白酶与底物浓度比1∶10,底物质量浓度60 g/L,pH 9.0,温度50℃的最佳酶解工艺条件下,所得大米硒代多肽的DPPH自由基清除率可达78.19%。因此,碱法提取是一种比较有效的大米含硒蛋白的提取方法,胰蛋白酶水解可获得较高抗氧化活性的大米硒代多肽。The extraction condition of Se-containing proteins, enzymatic preparation and antioxidant activity of seleno-peptides derived from Se-enriched rice endosperm was investigated. The results showed that maximum extractability was reached when rice endosperm was extracted at 40 ℃ for 3 h with using 1:20(w/v), 0.07 mol/L NaOH. Under this condition, the protein and selenium contents of Se-containing proteins were 80.18% and 0.583 mg/kg, respectively. Trypsin appears to be the most desirable for producing high antioxidant peptides from Se-containing rice proteins. The optimum enzymatic hydrolysis condition was selected at an enzyme than substrate concentration of 10%, an substrate concentration of 6%, pH 9.0 and a temperature of 50℃. Under the optimum condition, the DPPH free radical scavenging rates of prepared seleno-peptides was raised up to 78.19%. In conclusion, alkali extraction was one of the most effective methods to extract Se-containing proteins from rice endosperm, and seleno-peptides prepared by trypsin hydrolysis exhibited higher antioxidant activity.
