摘 要:南瓜籽不皂化物中含有植物甾醇,角鲨烯和生育酚等活性成分,是潜在的功能性食品,化妆品,药物的营养成分。以不皂化物的质量为指标,优化南瓜籽中不皂化物提取的酸解工艺条件,为南瓜籽的开发利用提供理论参数。利用单因素试验考察了酸解时间,酸解温度,液料比,HCl-乙醇浓度对不皂化物提取率的影响,在此基础上采用响应面法(RSM)中的Box-Behnken设计试验对酸解工艺进行优化。在酸解时间1.01 h,酸解温度60.54℃,液料比值30.31,HCl-乙醇浓度为5.45 mol/L时,不皂化物的理论提取率最高达1.177 98%。考虑实际情况,修正提取工艺条件为:时间1 h,温度60℃,液料比30∶1(V/m),HCl-乙醇浓度5.45 mol/L,经验证此条件下不皂化物得率为1.176 6%,较为接近理论值,说明采用Box-Behnken试验设计寻求的最佳工艺条件可行。Unsaponifiable matter from Pumpkin seed contain many bioactive compounds, such as phyotosterol, squalene and tocopherol, which potential as functional food, cosmetics, pharmaceutical ingredient. In the present study, based on the mass of unsaponifiable matter, acid hydrolysis extraction of unsaponifiable matter from Pumpkin seed was studied by adopting response surface methodology (RSM), aiming to provide theoretical parameters for its further exploitation. Four factors of reaction time, temperature, liquid/material ratio and HCl-ethanol concentration were investigated on the basis of single factor test, by Box-Behnken design and analyzing the response surface plots, the extraction parameters were optimized. Results showed that theoretical unsaponifiable matter yield of 1.177 98% was achieved at a reaction time 1.01 h, temperature of 60.54 ~C, liquid/ material ratio was 30.31 with 5.45 mol/L HCl-ethanol concentration. According to conditions of actual experiment, the optimum extraction parameters were revised, reaction time was 1 h, temperature was 60 ~C, liquid/material ratio was 30 : 1 (V/m), HC1- ethanol concentration was 5.45 mol/L, validated experiment resulted in a unsaponifiable matter yield of 1.176 6% which was close to the theoretic value, thus. Box-Behnken desima is a eood way to seek the ot^timal acid hydrolysis extraction conditions.
