摘  要:目的:研究沙枣可食部分乙醇提取物体内外抗肝肿瘤作用。方法:应用MTT法和移植性H22实体荷瘤小鼠,研究不同剂量提取物的体内体外抗肿瘤作用。结果:提取物对体外培养的人肝癌HepG2细胞增殖有显著的抑制作用,在培养前48 h内,抑制效果与浓度有明显的依赖关系,且提取物的浓度越大,抑制效果越好;在急性毒性试验中,当提取物最大剂量为27.4 g/kg时,对实验小鼠未表现出毒性;荷瘤小鼠模型研究表明,提取物对肝癌H22有显著的抑制作用,当剂量为500 mg/kg时对肿瘤的抑制效果最好,抑瘤率达53.37%,与阳性对照无显著差异;所有沙枣组小鼠的胸腺指数和脾脏指数均显著大于阴性对照组,说明提取物对荷瘤小鼠的免疫功能有显著的促进作用;对肿瘤组织的病理学切片观察表明,提取物具有较强的抗肿瘤作用。To investigate the antitumor effect of extracts of edible part from Elaeagnus Angustifolia L.(extracts),MTT colorimeter and H 22 cells-bearing mice models were evaluated.The results showed that the extracts significantly inhibited proliferation of the human hepatoma cell HepG2.During the first 48 h of culture stage,the inhibitory effect was in a does-dependent manner,the more high of the concentration,the more strong of the inhibitory effect.In the acute toxicity test,the highest does of 27.4 g/kg did not show toxicity to mice.The tumor-bearing mice model suggested that the extracts had significantly inhibitory effect on hepatoma cells H 22.When the dose of extracts was 500 mg/kg,tumor inhibition rate was 53.57%,which was higtest.However,the extracts could also increased the spleen index and thymus index,and displayed certain promotion action to the immune organ of tumor-bearing mice.The slice through the pathological observation of tumor tissue also indicated that extracts had good antitumor activity.The contets of polyphenols,flavonoids,proanthocyanidin and polysaccharides might be responsible for its antitumor activities.
