摘  要:为提高黑曲霉XTY产阿魏酸酯酶的酶活,采用响应面优化方法优化其液体深层发酵培养基。通过部分因子试验得出2个影响较显著的因素,分别为豆渣、蛋白胨添加量,确定其最大响应区域。结合中心组合试验及响应面分析方法,建立以阿魏酸酯酶比酶活为响应值的二次回归方程模型,获得最优的液体深层发酵培养基组成:2.26%豆粕,1.035%蛋白胨,0.3%KH2PO4,0.8%Na2HPO4.7H2O,0.01%NaC1和0.02%MgSO4.7H2O。在此发酵培养基下,黑曲霉XTY液体深层发酵产阿魏酸酯酶的比酶活达944.56 U/g(发酵5 d),较优化前提高了56%。其实验值与预测值基本相符,说明预测模型可靠性高,可用于阿魏酸酯酶液体深层发酵培养基的优化。In order to enhance the production of ferulic acid esterases by Aspergillus niger XTY,response surface methodology was applied to optimize the submerged cultivation medium.A fractional factorial design was used to investigate the main factors affecting ferulic acid esterases yield of Aspergillus niger XTY.The results showed that two factors played important roles in the production of ferulic acid esterases,which including bean dregs and peptone.Central composite design and response surface analysis were applied to establish the second-order regression equation model of the yield of ferulic acid esterases.Based on this model,the optimal fermentation conditions were obtained as follows:2.26% bean dregs,1.035% peptone,0.3%KH 2 PO 4,0.8%Na 2 HPO 4.7H 2 O,0.01%NaC1 and 0.02%MgSO 4.7H 2 O.Under the optimum conditions,the specific activity of ferulic acid esterases increased nearly 1.56 times to 944.56 U/g and the experimental values were consistent with the predicted values.These results suggest that the predicted model was reliable and suitable for the optimization of ferulic acid esterases.
