摘  要:研究小球藻多糖对α-淀粉酶有最大抑制作用时的提取工艺条件。采用水提醇沉法提取小球藻多糖,3,5-二硝基水杨酸比色法检测小球藻多糖在体外对α-淀粉酶活性的抑制作用,在单因素试验的基础上,利用响应面法对优化小球藻多糖的提取工艺,并采用二次回归旋转组合试验确定提取条件。结果表明,根据二次回归旋转组合试验得到小球藻多糖提取的最优条件为液料比87∶1(V∶m),提取温度85℃,提取时间43min,该条件下,小球藻多糖对α-淀粉酶的抑制率达79.67%,多糖得率为1.31%。Objective Studies on the optimal extraction technology of Chlorella polysaccharide(CPS)based on the activity ofα-amylase with maximum inhibition.Methods Chlorella polysaccharide was extracted by water extraction and alcohol precipitation.The inhibition rate of CPS onα-amylase activity was detected by DNS(3,5-dinitrosalicylate)method.The optimization strategy used was a series of one-factor-at-a-time investigations and subsequent response surface analysis based on a quadratic regression rotary combination design.Results The optimum extraction conditions were obtained as follows: Liquid-to-solid ratio of 87∶1(V ∶m),extraction temperature of 85℃and extraction times of 43min,and were found to be the optimum extraction technology for Chlorella polysaccharide based on its inhibition rate onα-amylase activity..Under these optimum conditions,the yield of polysaccharide was 1.31%,and theα-amylase inhibition rate reached 79.67%.
