产气肠杆菌α-乙酰乳酸脱羧酶基因的克隆及其表达

摘　　要：用 PCR方法扩增产气肠杆菌α-乙酰乳酸脱羧酶基因 (AL DC) ,得到 0 .78kb的 DNA片段 ,扩增片段重组到依赖 T4 RNA聚合酶的高效表达载体 p QE- 30中 ,在大肠杆菌中得到高效表达 ,酶活性可高达 180 U/m L发酵液 ,经稳定性测定 ,重组菌株在 37℃连续培养至 6The ALDC gene was cloned by PCR using Enterobactr aerogenes chromosome DNA as template and got 0.78 kb DNA fragment. The fragment was inserted directionally into a prokaryotic expression vector pQE 30 with high efficiency. The ALDC gene was overexpressed in E. coli M15, and the production of ALDC was over 180 U/mL, 100 times more than the parented strain. The recombinant strain was still stable after 67 generations at 37 ℃ without antibiotic selection.
