摘  要:羟脯氨酸是胶原蛋白的特有成分,通过测定乳制品水解液中的羟脯氨酸来鉴定乳制品中是否掺加了胶原水解蛋白。建立了高效液相色谱-质谱检测羟脯氨酸的方法,采用C8色谱柱,以乙腈-水为流动相,使用电喷雾离子源,正离子检测模式,选择离子方式扫描。结果表明,羟脯氨酸的检出限为1.5mg/kg,在质量浓度为0.1-12mg/L围内呈现良好线性,三个不同添加量的平均回收率为82.0%-91.5%,相对标准偏差为3.3%-8.5%。该方法流动相简单,无需对样品进行衍生,操作简便,定性准确,重现性好,可用于乳制品质量安全的监督检验。Hydrooxyproline is unique component of collagenous protein, this feaature can be used for the identifcafion of coUagenous protein of hydrooxyproline through the determination of dairy products to identify whether the addition of collagenous protein ingredients. This study estabhshed a high performance hquid chromatography-mass spectrometry(HPLC-MS)-method to determinate hydrooxyproline in dairy products which detected positive ionization followed by selected reaction monitoring, hydrooxyproline was separated by the column of C8 with the mobile phase of acetonitrile-water (95:5 ,V/V) . The results showed, when 10-500μg hydrooxyproline was fortified, the average recoveries were between 82.0% and 91.5%, the RSD was between 3.3% and 8.5%, the hmit of determination was 1.5 mg/kg. The calibration curve was linear over a range from 0.1 mg/L to 12 mg/L (R=0.9995).The method has the advantages of easy operation, good reproducibility and accuracy without derivatization. It can be used for the supervision and inspection of dairy quality and safety.
