摘  要:目的为提高食用油中黄曲霉毒素ELISA检测的精密度与准确度,对检测相关条件进行优化探讨。方法通过调整样品前处理以及免疫反应过程中一些实验参数,对比不同实验条件下实际样品测定值的RSD与回收率,取一定量样品提取液加入样品稀释液后按ELISA测定程序进行测定。结果在优化条件下样品加标回收率在75.5%~115.6%之间,同一个样品提取液做多孔平行其RSD小于8.5%。以超高效液相色谱串联质谱(UPLC—MS—MS)进行确认检测结果满意。结论优化后的检测条件有效地改善了检测过程中不确定性,样品检测结果的平行性和重复性较好,本实验对食用油中黄曲霉毒素B1的有关优化研究参数可以作为实际检测分析的参考,适用于实验室常规检测。Objective In order to improve the precision and accuracy of the determination of aflatoxin B1 in edible oil by ELISA, some relative detection conditions should be optimized and discussed. Methods After optimization of the pretreatment procedures for sample and some parameters in the immunoassay, the RSD and recovery rates were also compared. Results Un- der current optimized conditions, the recovery rate of sample was located within 75.5% - 115.6%. The RSD of the same sam- ple extraction solution detected by current modified ELISA immunoassay method was lower than 8.5%. Moreover, the ELISA experiments results were verified by UPLC - MS - MS test, and comparison results were also satisfied. Conclusion Under the optimized determination conditions, the performance was effectively improved, and the repeatability of sample detection was sat- isfied. The optimized parameters can be applied to practical determinations, moreover, the current methods will be apt to the common analysis of the aflatoxin B1 by ELISA for general laboratory.
