摘 要:目的观察补充蛋白质粉(protein powder,PP)对老龄大鼠抗氧化、抗衰老的作用,并探讨其可能机制。方法 12月龄雌性SD大鼠40只,随机分成对照组和蛋白质粉低(PPL,0.167 g/kg·bw)、中(PPM,0.833 g/kg·bw)、高(PPH,1.667 g/kg·bw)剂量组。对照组大鼠灌胃蒸馏水,另三组分别灌胃不同剂量的蛋白质粉混悬液,每日1次连续60d。实验结束时,心脏采血检测生化指标及抗氧化指标;取脑、肝、肾、脾脏等称重、检测总蛋白含量及脑脂褐质、脑单胺氧化酶(MAO)活性。结果补充蛋白质粉对老龄大鼠体重无显著影响,但实验组大鼠脑、肝脏、肾脏总蛋白浓度较对照组高。血清总蛋白、总铁结合力实验组显著高于对照组,血清总胆固醇、尿素氮显著低于对照组。血清超氧化物歧化酶(SOD)活性蛋白质粉中剂量组显著高于对照组,血清丙二醛(MDA)含量蛋白质粉低、中剂量组大鼠显著低于对照组。实验组大鼠MAO活性显著降低。结论适量的蛋白质粉补充能有效改善老龄大鼠营养状况并具有抗氧化、抗衰老作用。Objective To observe the anti-aging and antioxidant activities of protein powder (PP) in aged rats. Methods Forty aged female SD rats were randomly divided into 4 groups: control group, protein powder low (PPL), middle (PPM) and high-dose groups (PPH) (0.167, 0.833, 1.667 g/kg bw). The control group was treated with distilled water, while PP groups were i.g. administered with different doses of PP for 60 d. The blood biochemical and antioxidant indicators including superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) contents in serum were determined by biochemical methods. Total protein content in brain, liver, kidney and spleen were detected by Biuret method. Lipofusein (LF) contents and monoamine oxidase (MAO) activities in brain were detected by relative reagent kits. Results As compared to normal rats, the levels of total protein content (TP) and total iron binding capacity (TiBC) in serum were significantly increased in PP groups, but the levels of urea nitrogen (BUN) and total cholesterol (TC) in blood were significantly decreased. Compared with control group, the total protein content in brain, liver and kidney were increased in PP groups. While in PPM group, the activity of SOD in serum was significantly increased and the content of MDA significantly decreased as compared to control group. The activity of MAO in brain in PPM group was significantly decreased as compared to control group. Conclusion Supplement of appropriate amount of protein powder can improve the anti-aging and antioxidative activities in aged rats.
