摘 要:建立了葡萄酒中生物胺的一种柱前衍生反相高效液相色谱-紫外检测方法,以高氯酸提取葡萄酒中生物胺,丹磺酰氯为衍生剂,1,7-二氨基庚烷为定量内标,采用C18色谱柱分离,流动相为乙腈和水,梯度洗脱,流速为1mL/min,紫外检测波长254 nm。结果表明,色胺、腐胺、尸胺、组胺、酪胺、精胺和亚精胺在18 min内得到良好分离。在0.1~20 mg/L浓度范围内,各组分生物胺呈现良好线性相关(r>0.9)。在葡萄酒中添加生物胺混合标准溶液,平均回收率为95.63%~104.79%,相对标准偏差RSD小于10%。4种酒样的检测结果令人满意。采用柱前衍生高效液相色谱-紫外检测,满足了葡萄酒中生物胺检测的需要。A new method of pre-column derived reversed phase high performance liquid chromatography (HPLC) - ultraviolet detection had been developed for the determination of biogenic amines in grape wine. The process was as follows: biogenic amine in grape wine extracted by per- chlorate, dansyl chloride used as the derivative agent, 1, 7 - diamino heptane as quantitative internal standard, C^8 column used for the separation, the mobile phase was acetonitrile and water, gradient elution, the velocity was 1 mL/min, and ultraviolet detection wavelength was 254 nm. The results suggested that tryptamine, putrescine, resin amine, histamine, tyramine, spermine and spermidine got good separation within 18 min. In the concentration range of 0.1 ~ 20 mg/L, biogenic amines in each component presented good linear correlations (r 〉 0.9). After the addition of bio- genic amine mixed standard solution in grape wine, the average recovery rate was 95.63 %~ 104.79 % and RSD was less than 10 %. The determi- nation results of four wine samples in this experiment were satisfactory. Accordingly, pre-column derivative HPLC - ultraviolet detection could be used for the determination ofbiogenic amine in grape wine in practice.
